Investigation of serum nitric oxide and malondialdehyde levels in cattle infected with Brucella abortus

چکیده
The objective of this study was to determine serum nitric oxide (NO) and malondialdehyde (MDA) levels in cattle infected with Brucella abortus (B. abortus). Study cohort was comprised of 30 cattle aged between 2-3 years; 20 infected with B. abortus and 10 healthy cattle. B. abortus infection was determined in the animals by Serum Agglutination Test and Multiplex PCR assay. Serum NO level was determined by enzymatic Greiss reaction and MDA level by Yoshioka method. Mean serum NO and MDA levels of cases with confirmed B. abortus were 58.20 ± 6.50 μmol/L and 2.25 ± 0.10 nmol/ml, respectively whereas those of the controls were 38.07 ± 2.40 μmol/L and 1.74 ± 0.25 nmol/ml, respectively. We found that serum NO and MDA levels in infected cattle were significantly higher than healthy control cattle (p < 0.05 and p < 0.01, respectively). In conclusion, the increase in NO level in cattle infected with B. abortus is a result of stimulation of NO synthesis in macrophages by bacterial lipopolysaccharides, and the increase in MDA level is a result of excessive radical production due to brucellosis acting upon membrane lipids and can be used as an indicator of tissue damage.

High Level Expression of Recombinant Ribosomal Protein (L7/L12) from Brucella abortus and Its Reaction with Infected Human Sera
Brucellosis, caused by Brucella spp., is an important zoonotic disease that causes abortion and infertility in cattle and undulant fever in humans. Various studies have examined cell-free native and recombinant proteins as candidate protective antigens in animal models. Among Brucella immunogenes, antigen based on ribosomal preparation has been widely investigated. In this study, the immunogeni...

Background: Using Brucella abortus Strain 19 (S19) to control bovine brucellosis is restricted due to induce antibodies to the O-side chain of the smooth lipopolysaccharide (LPS) which may be difficult to differentiate vaccinated and infected animals. Furthermore, it is virulent for humans and can induce abortion to cattle. Objectives: The aim of...

We examined the expression and activity of inducible nitric oxide synthase (iNOS) in both gamma interferon (IFN-gamma)-treated and untreated murine macrophages infected with the gram-negative bacterium Brucella suis. The bacteria were opsonized with a mouse serum containing specific antibrucella antibodies (ops-Brucella) or with a control nonimmune serum (c-Brucella). The involvement of the pro...
متن کاملThe Assessment of Cytokine and Antibody Responses to Recombinant 31kDa Brucella Cell-Surface Protein in Brucella Abortus Infected Mouse Model
Background & Objective: One of the most common diseases between zoonosis - especially in developing countries – is brucellosis. Identification of Brucella cell antigen combinations in terms of the amount and type of immune response in infected hosts, are important in vaccine design. 31kDa Brucella cell surface protein (BCSP31) is shared among all Brucellae. We aimed to define specific immune r...
